Approximately 75% of breast cancers express estrogen receptor (ER). Endocrine therapy targeting the ER pathway is effective, but resistance leading to disease progression and metastasis is common and clinically challenging. A better understanding of the mechanisms of endocrine resistance and metastasis is needed to develop new therapies and predictive biomarkers to improve patient outcome. FOXA1 is a protein that regulates ER activity on gene expression. We have recently shown that aberrant activation of FOXA1 signaling leads to endocrine resistance and metastasis.
