This application seeks to renew the CPRIT Core Facility Support Award (RP160732) for the UTHSA Cancer Genome Sequencing and Computation Core (UTHSA-CGSCC). During the last funding period, CGSCC has made a remarkable contribution to the cancer research community in San Antonio, a Hispanic majority city and the catchment base for all South Texas, including many premier biomedical research institutions. As the only publicly accessible next-generation sequencing and computation core facility in the area, we focus our resources in implementing new sequencing methodology, new assay development, and genomic applications in health disparity research, as well as leveraging resources acquired through CPRIT funding to obtain additional federal funds to upgrade our instruments. The success of the CGSCC is thoroughly documented in the renewal summary, providing services and support to 65 cancer researchers and 80 NCI/CPRIT research grants with total funding exceeding $120 million, and contributing to greater than 80 peer-reviewed scientific publications. With the renewal of this CPRIT award, we will introduce the exciting "third-generation sequencing" technology to our cancer research community, matched with enhanced data storage capability, to enable the detection of rare genetic variations at the single-molecule level from our unique patient population. We also propose to significantly enhance our single-cell library preparation capability up to million cells per run, and to introduce spatial transcriptome techniques that will allow researchers to analyze single-cell gene expression in a wider variety of tissue samples, to reveal the cellular and spatial organization of tumors. These cutting-edge technologies will facilitate new discoveries in translational and clinical cancer research to improve care for patients in south Texas and beyond, which is the long-term goal of CGSCC.
